Abstract
Hepatitis C virus infection is a major health and economic burden across the world. Traditional medicines are associated with low sustained virological response and some adverse effects. Inhibitors of NS5A demonstrate potent pangenotypic activity in vitro and in vivo. This review summarized the journey of discovery and development of NS5A inhibitors including the early chemotypes for which resistance maps to the NS5A protein, the first, second generation and other analogues of NS5A inhibitors. The mechanism of action and clinical trials were also described.
